1Protocol: Human ex vivo skin tissue was used in this study. After culturing the skin tissue for 2 days, the skin tissue was UV irradiated (30J/cm2 UVA and 50mJ/cm2 UVB) for 4 consecutive days. After each irradiation, fresh culture medium was replaced and samples were applied to the skin surface. The positive control (VC+VE) group was administered under liquid, while the sample group was administered on the surface. After 4 consecutive days of irradiation, the ex vivo skin tissue was cultured for another 3 days, during which no irradiation was performed and only sample administration was performed. After the end of treatment, skin samples were taken and then biomarker analysis was performed. Immunofluorescence (IF) was used to detect the expression of collagen I, collagen III, collagen IV, and collagen XVII, immunohistochemistry (IC) was used to detect the expression of elastin, Ki67, Victoria blue staining was used to detect the expression of elastic fiber, Masson staining was used to detect the expression of collagen fiber, and ELISA was used to detect the expression of MMP-1.
2Protocol: A 12-week single-center clinical trial was conducted in 82 women with Fitzpatrick skin type I-VI (52% sensitive skin), age 34-65 years, and mild to moderate wrinkles, fine lines, visible pores, and dermatologist-assessed skin roughness. After cleansing, P-TIOX was applied twice daily in the morning and evening, followed by a standard moisturizer and sunscreen (AM). Subjects were instructed to avoid topical facial applications for at least 2 days prior to baseline. Efficacy assessments were conducted at baseline, weeks 1, 2, 4, 8, and 12.
